Studies on the resolution of 2-amino-1-butanol


Abstract
In resolution process, 2-amino-1-butanol was reacted with L-glutamic acid, production a diastereomer.The resolution was carried out by appling solubility difference upon the organic solvents.
The results were as follows;
1) In production of a diastereomer through the reaction between 2-amino-1-butanol and L-glutamic acid,the amounts of water should be minimal to monophase of two reactants. Thus, water produced minor effects upon the resolution of diastereomer.
2) The most suitable solvent for the resolution of diastereomer was an absolute ethhyl alcohol (99.7%). In the reaction of 0.11mole of 2-amino-1-butanol and L-glutamic acid,maximal resolution occurred at -4¢ª¡­ -7¢ªC with 140ml of absolute ethyl alcohol.Thus the (+)-fome of diasteromer, (+)-1-hydroxybutane-2-ammoniumhydrogen-L-glutamate, was at first precipitated as white crystals,resulting over 91% resolution. Within the solution (-)-1-hydroxybutane-2-ammoniunmhydrogen-L-glutamate was left.
3) The overall yield of (+)-2-amino-1-butanol and (-)-2-amino -1-butanol from (+)-from and (-)-from of diastereomer respectively,was about 75%
